Wal-Mart limits ammo sales after demand surges

Wal-Mart has started to limit sales of ammunition to three boxes per customer per day due to limited supplies, a spokeswoman said on Wednesday.?

Sales of guns and ammunition have risen across the United States since the December 14 shooting at a Newtown, Connecticut elementary school.?

Walmart U.S., is the largest unit of Wal-Mart Stores Inc, which is the world's largest retailer and the largest U.S. gun seller. The Bentonville, Arkansas-based retailer had said that it works to strike the right balance between serving hunters and sportsmen and ensuring that it sells firearms responsibly.?

"In order to take care of as many customers as possible, starting Thursday, January 24, all ammunition sales were limited to three boxes per customer, per day as supply is limited at this time," spokeswoman Ashley Hardie told Reuters on Wednesday.?

Wal-Mart was a charter member of New York City Mayor Michael Bloomberg's coalition against illegal guns and adopted a 10-point code established by the Responsible Firearms Retailer Partnership that goes beyond what the law requires.?

Data released on Monday by the Federal Bureau of Investigation's National Instant Criminal Background Check System showed that nine of the top 10 days for firearms checks from November 30, 1998, to January 27, 2013, including December 14, 2012, occurred since Adam Lanza killed 20 children and six adults at Sandy Hook Elementary School and then shot and killed himself. Lanza also killed his mother before the school massacre.?

Wal-Mart's policy change was reported earlier by The City Wire, an Arkansas news outlet.

Background checks on center stage at?Senate hearing

Updated 3:17 p.m. -?Democrats looking to sustain public pressure for new gun laws in the wake of the Newtown shootings clashed Wednesday with Republicans and the National Rifle Association over universal background checks, a far less dramatic proposed change than an assault weapons ban or limits on high capacity magazines.

"My problem with background checks is you're never going to get criminals to go through universal background checks," Wayne LaPierre, CEO and chief lobbyist for the NRA, said at a Senate Judiciary Committee hearing on gun violence, the first since President Barack Obama laid out new measures to curb gun crime. "None of it makes any sense in the real world."

The obvious drama in the packed hearing room lasted over four hours, with passions running well beyond the normal staid congressional panel. The emotion was heightened by the presence of some major iconic figures in the battle over whether ? and how ? to tighten federal regulation of firearms.

LaPierre sat at the opposite end of the witness table from Mark Kelly, the husband of former Rep. Gabrielle Giffords. Critically wounded at a shooting in Tucson in 2011, Giffords opened the hearing with a dramatic plea, haltingly asking Congress to "do something to prevent gun violence."

"My wife would not have been sitting here today if we had stronger background checks," Kelly told the committee later in the hearing.?

Under current law, people can buy guns through a private seller without getting a background check. It's commonly referred to as the "gun show loophole." The Obama administration's proposal to close this loophole by requiring background checks for all sales of firearms dominated much of Wednesday's hearing.

The exchanges at the hearing illustrated the sharp political divide over changing the nation's gun laws ? and the difficulty in enacting any of the more dramatic new measures included in the package the White House is pushing, which includes an assault weapons ban and limits on high capacity magazines.

"The deaths in Newtown should not be used to put forward every gun control measure that has been floating around for years," said Sen. Charles Grassley, R-Iowa, the committee's ranking member.

"Emotion often leads to bad policies," said Sen. Ted Cruz, R-Texas, who called the 1994 assault weapons ban a "singularly ineffective piece of legislation."

Gabrielle Giffords' husband, retired astronaut and Navy Capt. Mark Kelly, tells the Senate Judiciary Committee that he and his wife are still gun owners and value the second amendment, but stresses that the right to own a firearm demands responsibility and urges lawmakers to revise existing gun control legislation.

Even Chairman Patrick Leahy, a Democrat from rural Vermont, did not explicitly endorse the assault weapons ban that Sen. Dianne Feinstein, D-Calif., introduced last week. But he did call for background checks, sharply challenging LaPierre on the subject.

The NRA's position on background checks is a switch from the organization's position 14 years ago. "We think it's reasonable to provide mandatory instant criminal background checks for every sale at every gun show. No loopholes anywhere for anyone," LaPierre told a congressional panel in 1999.

A place where there was some common ground: gun trafficking.?

?We may be able to work together to prevent straw purchasers from trafficking in guns,? Grassley said, a sentiment echoed by others on the panel.

The obvious legislative hurdles -- on display Wednesday -- help explain why Democrats are relying on a campaign-like strategy and a series of public events to try to ratchet up public demand for stricter regulations on firearms. Giffords' story makes her a compelling public advocate.

"Too many children are dying," she said Wednesday, breaking up the syllables during her testimony.

"It will be hard, but the time is now," said Giffords, who has embarked on an arduous recovery since she was shot in the head, affecting her speech. "You. Must. Act. Be bold. Be courageous. Americans are counting on you."

She walked into Wednesday's hearing, her husband holding her hand and carefully guiding her to her seat in front of the Senate panel.

She spoke for just over a minute; her husband helped her back out of the room.

"Gabby's gift for speech is a distant memory," Kelly said later. "She struggles to walk, and she is partially blind. Her right arm is completely paralyzed."

In trying to counter the emotional testimony, Republicans repeatedly praised Giffords? perseverance and focused on trying to raise doubts about whether the measures Democrats had proposed to combat gun violence would work. They insisted current gun laws?aren't?being prosecuted effectively.

?This discussion, I sit here and listen to it, and my reaction is how little it has to do with the problem of keeping our kids safe and how much it has to do with the decadelong, two decadelong, gun ban agenda when we don?t even enforce the laws on the books,? LaPierre said.

Wednesday's hearings were the first in a planned series of sessions on gun laws. Leahy said Wednesday that he plans to begin the process of crafting a gun package in his committee next month. With Obama and Vice President Joe Biden publicly making the case for new laws, gun control advocates expect any action to begin in the Senate; the Republican-controlled House of Representatives has shown little appetite for taking up the issue.

In the wake of Newtown, a recent NBC News/Wall Street Journal poll showed that 56 percent of Americans believe gun laws should be more strict. The survey showed just 7 percent believe gun restrictions should be less strict.

Senate Majority Leader Harry Reid, D-Nev., said Tuesday that he planned to bring gun legislation to the Senate floor -- though with an open process that could allow senators to make changes. Such a process would likely make it harder to pass the bill.

?It?s very clear that there?s going to be a bill brought out of the committee, brought to the Senate floor, and there will be an amendment process there,? Reid said. He added that senators would be allowed to ?bring up whatever amendments they want that deal with this issue.?

Team Maxxis/H&M?s David Haagsma topped the podium at the WORCS ATV season kick-off, while teammate Beau Baron took third place at the race, held January 26-27 in Peoria, Arizona.

Riding through thick mud for the entire race, Haagsma took the lead with a pass at the end of the 13th lap. He held the front spot through to the finish line, and took first place with a seven-second margin of victory. Baron, who sparred with another rider for the third place spot for much of the event, scored the fastest Pro lap time of the race at 6:29.113 minutes. Both Haagsma and Baron trusted Razr Xc rears and Razr2 fronts for Round 1, and the tires were more than a match for the sloppy conditions. ?It rained a lot,? said Haagsma. ?We had a mud race, and I had a horrible start, but my tires got me all the way back up to the front, and I took first!?

The Pro contest wasn?t Baron?s only shot at a Round 1 podium: he also took first place in the Side x Side Pro Production race on 27? Vipr tires. In the Pro Am ATV class, Maxxis rider Grant Mitchell took second place.

Mayor Bloomberg's proposed budget could cut up to 1,800 teaching jobs

Mayor Bloomberg gave his final budget address as mayor on Tuesday, proposing a $70.1 billion plan that slashes up to 1,800 teaching positions through attrition.

The proposed budget doesn?t raise taxes or threaten the jobs of any city workers, but the mayor is targeting cherished city services to save costs.

Among programs in danger are the same 20 fire companies that have faced closure nearly every year of Bloomberg?s mayoralty ? only to be saved by City Council during budget negotiations ? and day care and after-school programs for as many as 47,000 children.

?The financial plan presented today continues to protect critical services and foster economic growth, while also taking the responsible, budget-minded actions that have resulted in a more efficient city government,? Bloomberg said as he released the budget.

The FDNY could be forced to shutter 20 compnaies under Mayor Mike's plan.

The budget would raise parking meter rates, cut funding for child vision screenings and slash the number of recreation days prisoners get from seven to five.

But the most controversial element in mayor?s proposal is his plan to cut 1,800 teaching jobs ? a move he said he?s been forced to make by a hostile union that won?t reach a deal with him on teacher evaluations.

The city lost more than $400 million in state and federal aid this year when it failed to meet a Jan. 17 deadline set by Gov. Cuomo for districts to make evaluation agreements with their unions.

The city could lose hundreds of millions more if the evaluation deal remains elusive for months to come.

The direct result will be the elimination of 700 teaching positions through attrition this year, and another 1,100 starting in September, the mayor said.

This could have a serious impact on kids, parents and educators said.

Oakland 'Coliseum City' Receives More Funding: $1 Million Approved For Study (VIDEO)

Despite threats to abandon Oakland from both the Athletics and the Warriors, the Oakland-Alameda County Coliseum Authority agreed to spend an additional $1 million studying a proposal for a massive sports and entertainment complex on Friday.

The plan, dubbed "Coliseum City," was first introduced early last year, and may be Oakland's only hope for retaining its sports teams--all of which could leave by 2017.

"The fear is legitimate but we're working really hard on a plan that could eventually still have all three teams," said Oakland mayor Jean Quan to the San Francisco Chronicle.

The new project would be modeled after Los Angeles' LA Live, a 27-acre complex that includes the Staples Center, as well as restaurants, bars, shopping and hotels. The estimated cost is upwards of $1 billion, but according to city officials, the project would largely be privately financed.

So far, the Raiders is the only team that has expressed interest in pursuing future development at the site. According to the San Jose Mercury News, the team has agreed to help fund the study approved on Friday. If the plan moves forward, the Raiders will reportedly be the anchor tenant.

"Coliseum City is a game-changer, and Oakland is playing to win," said Quan at a press conference last year.

The city council approved $3.5 million for design work and an environmental review in March. The additional $1 million approved on Friday will fund site planning and studies on revenue potential and market demand.

While the new center could mean a boon for Oakland, some residents expressed frustration that the funds were not being used elsewhere, particularly in light of the Warriors' long-ago announced plans to move to San Francisco.

"There's a lot of people and a lot of programs that could use that money, as opposed to just doing a negative study on something that's gone," said Oakland resident Donald Edward to ABC News.

Indeed, Oakland has seen its share of financial struggles. The Chronicle reported that crime in the city has spiked significantly since the city council laid off more than 100 members of the police department in response to its 2010 budget deficit.

But Stanford economist Roger Noll told the Chronicle that although stadium building isn't always a wise investment for a city, including one in a larger development can significantly benefit both a team and a city.

"The current wave of how new facilities are built, you embed them in larger developments," he explained. "The teams' benefit is indirect."

"It's not about the sports stadium," said Mayor Quan to the Chronicle. "It's about how much more profit (can be made) off of the development around the sports teams. The area around the Coliseum is one of the largest areas of undeveloped land in a major urban area, so there's a lot of potential. And it would mean more than 30,000 new jobs."

A barge carrying 80,000 gallons of oil hit a railroad bridge in Vicksburg, Miss., on Sunday, spilling light crude into the Mississippi River and closing the waterway for eight miles in each direction, the Coast Guard said. A second barge was damaged.

Investigators did not know how much had spilled, but an oily sheen was reported as far as three miles downriver of Vicksburg after the 1:12 a.m. incident, said Lt. Ryan Gomez of the Coast Guard's office in Memphis, Tenn.

Authorities were still trying to determine the source of the leak, but it appeared to be coming from one or two tanks located at the stern of the first barge, Gomez said. He said there was no indication that any oil was leaking from the second vessel, and said it was still unclear whether the second barge also hit the bridge or was damaged through a collision with the first.

"Investigators are still trying to figure out what happened," he said.

United States Environmental Services, a response-and-remediation company, was working to contain the oil with booms before collecting it and transferring it to one of the barge's undamaged tanks, then ultimately to a separate barge, Gomez said. He could not say how long the river would remain closed in the area. Five northbound and two southbound vessels were waiting to pass, he said.

"It's still considered an active leak," Gomez said. "We don't have an estimate or accurate amount of what was released."

Railroad traffic was allowed to continue after the bridge was found safe for trains, Petty Officer Carlos Vega said.

The barges are owned by Third Coast Towing LLC, Gomez said. According to a website listed under that name, the company is located in Corpus Christi, Texas. No one answered the telephone at the company Sunday night.

Both vessels were being pushed by the tugboat Nature's Way Endeavor. The website for Nature's Way Marine LLC of Theodore, Ala., identifies the vessel as a 3,000-horsepower, 90-foot-long boat, making it the largest and highest-powered of the company's five tugs. It was built in 1974 and underwent a complete rebuild in 2011, according to the company.

A company manager referred calls to the Coast Guard command center at Vicksburg.

The last time an oil spill closed a portion of the lower Mississippi River, it was for less than a day last February after an oil barge and a construction barge collided, spilling less than 10,000 gallons of oil. In 2008, a fuel barge collided with a tanker and broke in half, dumping 283,000 gallons of heavy crude into the waterway, and closing the river for six days.

The oil sheen from Sunday's incident was unlikely to pose a threat to the Gulf of Mexico, located 344 river miles south of Vicksburg.

Residents and businesses in Gulf Coast states are still recovering from the April 2010 explosion of the Deepwater Horizon drilling rig, which killed 11 workers and spewed more than 200 million gallons of oil into the Gulf.

Deadly smoke, lone blocked exit: 230 die in Brazil

SANTA MARIA, Brazil (AP) ? A fast-moving fire roared through a crowded nightclub in southern Brazil early Sunday, within seconds filling the space with flames and a thick, toxic smoke that killed more than 230 panicked partygoers who gasped for breath and fought in a stampede to escape.

It appeared to be the world's deadliest nightclub fire in more than a decade.

Firefighters responding to the blaze at first had trouble getting inside the Kiss nightclub because bodies partially blocked the club's entryway.

Witnesses said a flare or firework lit by band members started the blaze in Santa Maria, a university city of about 260,000 people. Officials at a news conference said the cause was still under investigation ? though police inspector Sandro Meinerz told the Agencia Estado news agency the band was to blame for a pyrotechnics show and that manslaughter charges could be filed.

Television images showed black smoke billowing out of the Kiss nightclub as shirtless young men who had attended a university party joined firefighters using axes and sledgehammers to pound at windows and hot-pink exterior walls to free those trapped inside.

Bodies of the dead and injured were strewn in the street and panicked screams filled the air as medics tried to help. There was little to be done; officials said most of those who died were suffocated by smoke within minutes.

Within hours a community gym was a horror scene, with body after body lined up on the floor, partially covered with black plastic as family members identified kin.

Outside the gym police held up personal objects ? a black purse, a blue high-heeled shoe ? as people seeking information on loved ones looked crowded around, hoping not to recognize anything being shown them.

Guido Pedroso Melo, commander of the city's fire department, told the O Globo newspaper that firefighters had a hard time getting inside the club because "there was a barrier of bodies blocking the entrance."

Teenagers sprinted from the scene after the fire began, desperately seeking help. Others carried injured and burned friends away in their arms. Many of the victims were under 20 years old, including some minors.

"There was so much smoke and fire, it was complete panic, and it took a long time for people to get out, there were so many dead," survivor Luana Santos Silva told the Globo TV network.

The fire spread so fast inside the packed club that firefighters and ambulances could do little to stop it, Silva said.

Another survivor, Michele Pereira, told the Folha de S. Paulo newspaper that she was near the stage when members of the band lit flares that started the conflagration.

"The band that was onstage began to use flares and, suddenly, they stopped the show and pointed them upward," she said. "At that point, the ceiling caught fire. It was really weak, but in a matter of seconds it spread."

Guitarist Rodrigo Martins told Radio Gaucha that the band, Gurizada Fandangueira, started playing at 2:15 a.m. "and we had played around five songs when I looked up and noticed the roof was burning"

"It might have happened because of the Sputnik, the machine we use to create a luminous effect with sparks. It's harmless, we never had any trouble with it.

"When the fire started, a guard passed us a fire extinguisher, the singer tried to use it but it wasn't working"

He confirmed that accordion player Danilo Jacques, 28, died, while the five other members made it out safely.

Police Maj. Cleberson Braida Bastianello said by telephone that the toll had risen to 233 with the death of a hospitalized victim. He said earlier that the death toll was likely made worse because the nightclub appeared to have just one exit through which patrons could exit.

Survivors said security guards briefly tried to block people from exiting the club. Brazilian bars routinely make patrons pay their entire tab at the end of the night before they are allowed to leave.

Officials earlier counted 232 bodies that had been brought for identification to a gymnasium in Santa Maria, which is located at the southern tip of Brazil, near the borders with Argentina and Uruguay.

Federal Health Minister Alexandre Padhilha told a news conference that most of the 117 people treated in hospitals had been poisoned by gases they breathed during the fire. Only a few suffered serious burns, he said.

Brazil President Dilma Rousseff arrived to visit the injured after cutting short her trip to a Latin American-European summit in Chile.

"It is a tragedy for all of us," Rousseff said.

Most of the dead apparently were asphyxiated, according to Dr. Paulo Afonso Beltrame, a professor at the medical school of the Federal University of Santa Maria who went to the city's Caridade Hospital to help victims.

Beltrame said he was told the club had been filled far beyond its capacity during a party for students at the university's agronomy department.

Survivors, police and firefighters gave the same account of a band member setting the ceiling's soundproofing ablaze, he said.

"Large amounts of toxic smoke quickly filled the room, and I would say that at least 90 percent of the victims died of asphyxiation," Beltrame told The Associated Press by telephone.

"The toxic smoke made people lose their sense of direction so they were unable to find their way to the exit. At least 50 bodies were found inside a bathroom. Apparently they confused the bathroom door with the exit door."

In the hospital, the doctor "saw desperate friends and relatives walking and running down the corridors looking for information," he said, calling it "one of the saddest scenes I have ever witnessed."

Rodrigo Moura, identified by the newspaper Diario de Santa Maria as a security guard at the club, said it was at its maximum capacity of between 1,000 and 2,000, and partygoers were pushing and shoving to escape.

Santa Maria Mayor Cezar Schirmer declared a 30-day mourning period, and Tarso Genro, the governor of the southern state of Rio Grande do Sul, said officials were investigating the cause of the disaster.

The blaze was the deadliest in Brazil since at least 1961, when a fire that swept through a circus killed 503 people in Niteroi, Rio de Janeiro.

Sunday's fire also appeared to be the worst at a nightclub since December 2000, when a welding accident reportedly set off a fire at a club in Luoyang, China, killing 309.

In 2004, at least 194 people died in a fire at an overcrowded nightclub in Buenos Aires, Argentina. Seven members of a band were sentenced to prison for starting the flames.

A blaze at the Lame Horse nightclub in Perm, Russia, killed 152 people in December 2009 after an indoor fireworks display ignited a plastic ceiling decorated with branches.

Similar circumstances led to a 2003 nightclub fire that killed 100 people in the United States. Pyrotechnics used as a stage prop by the 1980s rock band Great White set ablaze cheap soundproofing foam on the walls and ceiling of a Rhode Island music venue.

The band performing in Santa Maria, Gurizada Fandangueira, plays a driving mixture of local Brazilian country music styles. Guitarist Martin told Radio Gaucha the musicians are already seeing hostile messages.

"People on the social networks are saying we have to pay for what happened," he said. "I'm afraid there could be retaliation".

An Army wife, denied full membership at a Fort Bragg spouses club because she's married to a female Army officer, was named Fort Bragg?s 2013 ?spouse of the year? Friday after an annual, national vote that spanned nominees from all military branches, NBC News has learned.

In the online election held Tuesday, Ashley Broadway captured the Fort Bragg vote ?by a country mile,? said Babette Maxwell, founder of Military Spouse magazine and the Military Spouse of the Year award. Ballot totals were not revealed. As one of the 154 base-level winners, Broadway now is eligible to be nominated for Army ?spouse of the year.?

?A lot of people who voted never me met or talked to me or knew me from Adam. I know it was a statement to the Obama Administration, to Secretary (of Defense Leon) Panetta, to Senator (Chuck) Hagel ? if he is confirmed (as defense secretary) ? to the Pentagon and, really, to America that, yes, she is a military spouse and she needs to be recognized,? Broadway told NBC News.


?There are things the government can do right now to make life a hell of a lot easier than what it is currently for those who are in same-sex marriages in the military,? she added. ?It was a lot of people saying, ?Enough?s enough.? ?

Broadway?s rejection from the Fort Bragg officers? spouses club sparked the U.S. Marine Corps to issue on Jan. 9 a pro-gay, branch-wide directive. On Jan. 16, her bid drew the Pentagon?s attention. The next day, the?on-base spouses club offered Broadway a "special guest membership" ? an invitation she declined, calling it ?extremely demeaning.??

Broadway married her 15-year companion, Lt. Col. Heather Mack, in November ? their first chance to hold a formal ceremony after?the 2011 repeal of ?Don?t Ask Don?t Tell,??the policy that kept gays from openly serving in the military. The couple has a 2-year-old son and Mack gave birth to their second child, a daughter, on Tuesday.?

?People got one vote per email address ? one ballot for the person you wanted to represent you. I think people would be unwilling to, quote-unquote, throw their vote away on simply doing what was popular,? Maxwell said. ?There was a significant amount of meaning in what they were doing when they voted for Ashley.

?Removing her a bit from the press and recognition she?s received the last few months, Ashley???more importantly???has a platform to benefit a large number of spouses, and that?s what people want to see happen,? Maxwell added. ?The winners are chosen based on their merits, their accomplishments and what they intend to do for the community in the year to come.?

Broadway has volunteered to tutor soldiers? children in reading, briefed inbound Army families on local school districts, and helped transferring soldiers with housing-location decisions.

?When I was denied membership, I asked to speak to the club?s board. I was convinced that if they?d just sit down with me for half an hour, if I could talk to them about what I?ve been doing, what I?ll be doing in the future, they would see what an asset I would be to the group,? Broadway said.

The meeting was not granted.

?That was the most frustrating thing,? she said.

The club?s board maintains Broadway was never rejected because ?a formal application was never filed,? and that she simply had inquired about the eligibility of a same-sex spouse and was told the club would need ?time to look at the issue.??

Online voting for the next round of the 2013 Armed Forces Insurance Military Spouse of the Year???the branch level???will take place Feb. 5. The overall winner, elected from the branch finalists, will be revealed May 9.

"I never thought in a million years I would be the one to advance the cause. If that?s what it?s going to take to get attention for all the military same-sex spouses, then so be it," Broadway said. "But I do take this (Bragg 'spouse of the year' award) very seriously. And we'll see where it goes from here."?

Canada's Flaherty vows tight spending to meet budget target

DAVOS, Switzerland (Reuters) - Canada's federal government must be very tough on spending or it won't be able to balance the budget as promised, Finance Minister Jim Flaherty said in an interview.

Even faced with weaker-than-expected growth, which could dent revenues, Flaherty upheld Ottawa's pledge to return to fiscal balance by 2015.

"On the spending side we're being really tough and I'm being really tough and we need to be really tough or I won't be able to balance the budget, which we're going to do by 2015," he told Reuters at the World Economic Forum meeting in Davos, Switzerland.

Flaherty is expected to present his next budget in March.

Figures for the first eight months of the fiscal year showed the deficit narrowing gradually, and Ottawa expects a relatively small deficit in the 2012-13 fiscal year of C$25 billion ($24.79 billion).

The lowest inflation rate in three years in Canada also makes the job of balancing the budget more difficult.

The level of nominal gross domestic product, which is GDP plus inflation, directly affects tax revenues and muted inflation means the revenues will be lower than they would be otherwise.

(AP) ? Top international financial officials wrapped up the World Economic Forum in the Swiss resort of Davos warning that much needs to be done to stabilize the world economy.

Despite relief that the euro, for all its struggles, remains intact and the U.S. has so far managed to get through a crucial budget hurdle, the International Monetary Fund's managing director Christine Lagarde urged world leaders to "not relax."

Lagarde said Saturday that the 17 European Union countries that use the euro have to follow through on steps to keep the troubles at banks from burdening governments.

And she said U.S. officials have to "indicate very promptly" how they're going to deal with their ongoing budget dispute between President Barack Obama and Republicans in Congress.

New method identifies genes that can predict prognoses of cancer patients

Jan. 25, 2013 ? In recent years, it has been thought that select sets of genes might reveal cancer patients' prognoses. However, a study published last year examining breast cancer cases found that most of these "prognostic signatures" were no more accurate than random gene sets in determining cancer prognoses. While many saw this as a disappointment, investigators at Beth Israel Deaconess Medical Center (BIDMC), the Dana-Farber Cancer Institute, and the Institut de Recherches Cliniques de Montr?al (IRCM) saw this as an opportunity to design a new method to identify gene sets that could yield more significant prognostic value.

Led by Andrew Beck, MD, Director of the Molecular Epidemiology Research Laboratory at BIDMC, the team has developed SAPS (Significance Analysis of Prognostic Signatures), a new algorithm that makes use of three specific criteria to more accurately identify prognostic signatures associated with patient survival.

Their results, the largest analysis of its kind ever performed, are reported in the January 24 on-line issue of the journal PLOS Computational Biology.

"SAPS makes use of three specific criteria," explains Beck, who is also an Assistant Professor of Pathology at Harvard Medical School. "First, the gene set must be enriched for genes that are associated with survival. In addition, the gene set must separate patients into groups that show survival differences. Lastly, it must also perform significantly better than sets of random genes at these tasks."

In the new study, the scientific team applied the SAPS algorithm to gene expression profiling data from the study's senior author Benjamin Haibe-Kains, PhD, Director of the Bioinformatics and Computational Genomics Laboratory at IRCM and an Assistant Research Professor at the University of Montreal. The first collection of data was obtained from 19 published breast cancer studies (including approximately 3800 patients), and the second included 12 published gene expression profiling studies in ovarian cancer (including data from approximately 1700 patients).

When the investigators used SAPS to analyze these previously identified prognostic signatures in breast and ovarian cancer, they found that only a small subset of the signatures that were considered statistically significant by standard measurements also achieved statistical significance when evaluated by SAPS.

"Our work shows that when using prognostic associations to identify biological signatures that drive cancer progression, it is important to not rely solely on a gene set's association with patient survival," says Beck. "A gene set may appear to be important based on its survival association, when in reality it does not perform significantly better than random genes. This can be a serious problem, as it can lead to false conclusions regarding the biological and clinical significance of a gene set."

By using SAPS, Beck and his colleagues found that they could overcome this problem. "The SAPS procedure ensures that a significant prognostic gene set is not only associated with patient survival but also performs significantly better than random gene sets," says Beck. His team revealed new prognostic signatures in subtypes of breast cancer and ovarian cancer and demonstrated a striking similarity between signatures in estrogen receptor negative breast cancer and ovarian cancer, suggesting new shared therapeutic targets for these aggressive malignancies.

The findings also indicate that the prognostic signatures identified with SAPS will not only help predict patient outcomes but might also help in the development of new anti-cancer drugs. "We hope that markers identified in our analysis will provide new insights into the biological pathways driving cancer progression in breast and ovarian cancer subtypes, and will one day lead to improvements in targeted diagnostics and therapeutics," says Beck. "We also hope the method proves widely useful to other researchers." To that end, the team would like to create a web-accessible tool to enable investigators with little knowledge of statistical software and programming to identify gene sets significantly associated with patient outcomes in different diseases.

"We also plan to soon release a software package, which includes all the code and corresponding documentation of our analysis pipeline," adds Haibe-Kains. "This will allow others to fully reproduce our results while enabling the bioinformatics and computational biology communities to take over and potentially adapt and improve our pipeline to address important new issues in biomedicine."

Beck and his collaborators are currently working to further validate the prognostic signatures they identified in breast and ovarian cancers, with the hopes of bringing them closer to the clinic through the development of new diagnostics and treatments. "We are also extending our approach to other common cancers that lack robust prognostic signatures," he notes.

New Genetic Twist: 4-Stranded DNA Lurks in Human Cells

Sixty years after scientists described the chemical code of life ? an interweaving double helix called DNA ? researchers have found four-stranded DNA is also lurking in human cells.

The odd structures are called G-quadruplexes because they form in regions of deoxyribonucleic acid (DNA) that are full of guanine, one of the DNA molecule's four building blocks, with the others being adenine, cytosine, thymine. The structure comprises four guanines held together by a type of hydrogen bonding to form a sort of squarelike shape. (The DNA molecule is itself a double strand held together by these building blocks and wrapped together like a helix.)

The new visualization of the G-quadruplex is detailed this week in the journal Nature Chemistry.

"I think this paper is important in showing directly the existence of this structure in vivo in the human genome, but it is not completely unexpected," said Hans-Joachim Lipps, of the University of Witten in Germany, who was not involved in the study. [See Images of the 4-Stranded DNA]

Scientists had shown in the past that such quadruplex DNA could form in test tubes and had even been found in the cells of ciliated protozoa, or single-celled organisms with hairlike appendages. Also there were hints of its existence in human cells, though no direct proof, Lipps said.

But scientists still didn't have concrete evidence for its existence in the human genome. In the new study, researchers, including chemist Shankar Balasubramanian, of the University of Cambridge and Cambridge Research Institute, crafted antibody proteins specifically for this type of DNA. The proteins were marked with a fluorescent chemical, so when they hooked up to areas in the human genome packed with G-quadruplexes, they lit up.

They also found a spike in quadruplexes during the s-phase of the cell cycle, or the phase when DNA replicates just before the cell divides.

As such, the researchers think the four-stranded DNA could be a target for personalized medicine in the future. If they could block these odd ducks perhaps they could stop the rapid cell division of cancer cells.

"We are seeing links between trapping the quadruplexes with molecules and the ability to stop cells dividing, which is hugely exciting," Balasubramanian said in a statement.

The finding "is certainly a technical (not scientific) breakthrough in designing antibodies sensitive enough to demonstrate this structure in vivo in the human genome," Lipps wrote.

Lipps and his colleagues had suggested previously these structures regulate basic biological mechanisms, such as the replication of DNA.

"What makes me personally very happy about this work is that it again demonstrates that mechanisms first described in ciliated protozoa hold also true for other organisms up to human, demonstrating the strength of this model organism," wrote Lipps wrote.

The team still has several questions about quadruplexes, such as how the structures operate. "One thought is that these quadruplex structures might be a bit of a nuisance during DNA replication ? like knots or tangles that form," Balasubramanian said.

East Africa economies set to benefit from energy prospects

JOHANNESBURG (Reuters) - Booming oil and gas investment in East Africa will sustain robust economic growth in Uganda and Tanzania, despite the corruption and power failures blighting the nations, a Reuters poll found on Thursday.

Tanzania, the region's no.2 economy, could see its economy expand around 7.0 percent this year from an estimated 6.6 percent in 2012, before growing another 7.1 percent next year, the survey showed.

"We are forecasting Tanzania to continue to outperform the region in terms of economic growth in 2013 on the back of continued investment in the energy sector and infrastructure," said Mark Bohlund, economist at IHS Global Insight.

Still, like neighbouring Uganda and Kenya, Tanzania frequently suffers from blackouts, and the International Monetary Fund (IMF) has warned it needs to limit power outages if it wants to maintain buoyant growth.

Uganda, the third biggest economic power in the region, could see growth of 5.6 percent this year from 4.2 percent last year, with a further acceleration to 6.5 percent in 2014.

That's a fairly similar outlook to Kenya, East Africa's largest economy according to a Reuters poll in November.

Analysts do not expect power outages in Tanzania and the cloud of corruption hanging over Uganda's government to deter further investment into East Africa's untapped energy deposits.

For Uganda, major western donors including Britain, its biggest bilateral source of aid, suspended their financial support to the country toward the end of last year after allegations of $13 million in stolen aid.

"In any case, continued exploration and investment in the natural resource sector should more than offset the effect of the aid freeze," said Shilan Shah, Africa analyst at Capital economics.

The government is hopeful that foreign donors will restore aid, which makes up a quarter of the nation's budget, after it pledged to refund stolen and embezzled funds.

A drought in 2011 prompted an inflation surge through East Africa, but better rains and tighter monetary policy stance by the central banks are subduing inflation, albeit slowly.

Ugandan inflation should ease from an average 14.6 percent last year to around 6.8 percent in 2013, and reaching 7.9 percent next year.

The Central Bank of Uganda left its benchmark lending rate at 12 percent at the beginning of this month - ending a run of growth-boosting cuts that began last June when the base rate was 21 percent.

Tanzanian inflation should also fall into single digits this year after averaging an estimated 16 percent in 2012. The poll showed it easing to 9.6 percent this year, and then to 7.8 percent next year.

"We are forecasting inflationary pressures to be relatively contained in 2013 on the back of sluggish economic activity," added Bohlund from IHS Global Insight.

"But (they) remain vulnerable to increasing food and energy prices as well as a weaker shilling."

North Charlotte fire forces family out of home | CharlotteObserver.com

Charlotte firefighters were called to a house fire on North Johnson Street just before 8 p.m. Friday night.

Firefighters were able to control the fire after about 15 minutes but the home suffered severe damage inside.

Nobody was hurt but the family of three have been displaced for the night. The Red Cross is helping them with shelter.

Investigators have not yet determined the cause of the fire.
